Use any of the following configuration

acl adclick1 dstdomain .doubleclick.net
acl adclick2 dstdomain .valueclick.net
acl adclick3 dstdomain .falkag.net
http_access deny adclick1
http_access deny adclick2
http_access deny adclick3

Or

acl adclick1 dstdomain .doubleclick.net .valueclick.net .valueclick.net
http_access deny adclick1

After configuring this, you can give reload the configuration file without
restarting

/PATH/TO/squid -k reconfigure
